Precision STING delivery could help convert immune-desert tumors into inflamed tumors

A new review highlights the cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) pathway as a central switch that can convert immunologically "cold" tumors into "hot," inflamed tumors and improve immunotherapy responses. The authors synthesize how acute activation of this innate immune sensor promotes antigen presentation, T-cell priming, and type I interferon (IFN-I) signaling, while chronic or mistimed activation can drive immunosuppression, metastasis, and treatment resistance. The review maps next-generation STING-targeted agonists and delivery platforms--including antibody-drug conjugates (ADCs), immune-stimulating antibody conjugates (ISACs), nanoparticles, bacterial vectors, and exosomes--to distinct tumor microenvironment (TME) phenotypes. It proposes a strategic framework for rational combination therapies and biomarker-driven patient selection.
